THE EFFECT OF ROSA DAMASCENA EXTRACT ACTIVITY ON ANGIOTENSIN CONVERTING ENZYM AND RENAL FUNCTION IN WISTAR RATS INDUCED HIGH DIET OF NATRIUM

ABSTRAK
Background: The prevalence of hypertension still keeps increasing in accordance with the changing of lifestyle. Hypertension is known as ‘the silent killer’ because it can appear suddenly and cause various complications which eventually results to death. Complication in hypertension si damage kidney. One of the causes of hypertension is the excess intake of natrium so that it causes the imbalance of liquid inside the body managed by the mechanism of Renin Angiotensin Aldosterone System (RAAS). The increasing activity of Angiotensin Converting Enzyme (ACE) in body can cause hypertension through the mechanism of RAAS. The activity of ACE can be blocked by ACE Inhibitor (ACE-I), which is one of the basic treatments of hypertension.
Objective: The purpose of this research is to know the effect of the administering of Rosa damascene extract (RE) to the activity of ACE, renal function (urea, creatinine) and renal histopathology in Wistar rat with natrium diet.
Methods: This research is experimental research with post-tests only control group design. The subject of research is male Wistar rat with 150-200 gram in weight, divided into five groups, they are: P0 = control group; P1 = NaCl 8% + RE 500 mg/kgBW; P2 = NaCl 0,35% + RE 500 mg/kgBW; P3 = NaCl 8% and P4 = NaCl 0,35% for 28 days. After 28 days-treatment, anasthesia was done to the rat with Ketamin-Xylazine combination. Rat blood was taken from the heart for ACE by ELISA method, uremic, creatinine by spectrophotometer.
Results: The used data analysis is One Way Anova test to the difference among the treatment groups. it was found the difference of ACE activity, urea, creatinine serum in Wistar rat which was before given natrium diet with value of p < 0.05. Rose flower extract (Rosa damascene) has been proved to be able to decrease the activity of ACE, urea, creatinine serum in Wistar rat which was before given natrium diet with value of p < 0,05. Rosa damascene ethanol extract is able to provide a nephroprotection effect.
Conclusion: Rose flower extract (Rosa damascene) had activity against ACE inhibition, as well as protection of renal function in rats induced by a high sodium diet.
